Hello everyone,
 
After a week of experimenting with bremelanotide (PT-141), I'm able to give an update.  
 
1st try: me and girlfriend both took 2 mg sub-q.  
Girlfriend: side effects none. Libido: no effect.  
Me: side effects: flush, naussea, headache (bad one!), fatigue, profuse sweating, extreme heavy feelings in legs - for about 30 hours afterwards. Libido: no effect.  
 
Conclusion: no effect on libido. Further mode of action: no more sub-q injections for me, side effects are too strong. Now trying internasally.  
 
2nd try: me only: 5 mgs mixed with 2 mls of 'water for injections' and taken internasally with atomized spray. Side effects: flush.  
1 hour later: libido: no effect BUT when stimulated good, quick & pleasant orgasm (2x).  
 
3rd try: me only: 10 mgs nasally, same method of application as above. Side effect: stronger Bremelanotide flush.
1 hour later: no effect on libido, BUT when stimulated, 2x orgasm, not very strong, but pleasant nontheless.
 
4th try: me only: 20 mgs nasally, same method of application. Side effect: strong Bremelanotide flush which lasted for about 1 hour.  
1 hour later: shock type of feeling, extreme diarrea, low blood pressure, extreme cold and shivery for about 30 minutes. Libido: no sexual fantasies, desire or anything like that (unfortunately).  
7 hours later: when stimulated: 1 orgasm with girlfriend, not very strong.  
5 hours later: masturbating: 1 good strong orgasm, took a while to achieve though.  
 
Eventhough there does seem to be an effect on orgasms, I'm dissapointed with the overal effect. I thought Bremelanotide was reputed to enhance libido too (sexual thoughts and feelings and such).  
 
I'm wondering what's wrong. I.e. the shipment was not delivered with ice packs... could this be the clue? By the time I received the parcel it had been in transit for at least 10 days.  
 
Or - is it not the true stuff I received? May be just a cheap knock off?  
Or - is the 'water for injections' not the right vehicle?  
 
This is all very confusing to me.  
 
May be I should stick to the sub-q method and try a higher dose, but I've turned chicken since the last time I felt sick for about 30 hours.  
 
Any clues, anyone?

Maria - please let us know the reconstitution protocol you used.  I read a thread a couple of days ago that it is important to strictly follow the procedure as outlined in that thread.  Otherwise - poor results.

Maria - please search for: room temp. , room temperature , reconstitute
 
Here is some info and links I've found on the web:
 
"Amino acids such as Cysteine, Methionine or Tryptophan are susceptible to oxidation and should be dissolved or stored in oxygen free water."
 
"Peptide Storage and Handling
 
All Princeton Peptides are supplied lyophilized under argon. For maximum stability the lyophilized peptide should be stored at -20 °C. Upon removal from the freezer, the peptide vial should be brought up to room temperature prior to opening and weighing to avoid moisture condensation.
 
Attempt to re-suspend the peptide in sterile distilled deionized water. If the peptide is insoluble, addition of a mild alkali such as 0.1% NH4OH can increase solubility of acidic peptides. On the contrary, mild acids such as 0.1% TFA can help solubilize basic peptides.
 
For peptides which contain methionine, cystenic or tryptophan residues, oxidation may generate impurities with or without concomitant biological inactivation. To avoid this, peptides containing these amino acids should be dissolved in oxygen-free solvents."

Questions that arise (at least for me) are as follows:
 
Q1:  Is bacteriostatic water the best dilutant for bremelanotide (PT-141) considering its toxcity (however slight for adults)?  
Q2:  Is bacteriosatic water deionized?
Q3:  Does the benzyl alcohol in bacteriostatic water modify the chemistry (even slightly) of bremelanotide (PT-141)?
Q3:  Is sterile water deionized?
Q4:  Is deionized water sterile?
 
I will research these questions further.  Those much more familar with peptides please feel free to voice your opinions.
 
 As for me I think I will lean towards sterile water or deionized water as a dilutant for bremelanotide (PT-141).

I snipped the following out of a post by TheGame46 over on SuperiorFitness:  
 
 "For  IGF you use an acetic acid solution. If one was not made available to you you can make the solution using 7 parts distilled water and 1 part vinegar from the grocery store. You must  filter this through a sterile syring  filter before use however.
 
For MGF use Bacteriostatic Water BW.
 
For HGH Fragments and GHRP's also use BW.
 
When reconstituting you are going to add the liquid to th vial containing the powder is a slow controled manner with the vial tilted so that the liquid trickles out of the needles and rolls down the side of the vial. Do not squirt it directly into the peptide b/c this may damage it.

Now he isn't talking about bremelanotide (PT-141), but if bremelanotide (PT-141) is as delicate as it seems, perhaps we need to make sure we reconstitute it as delicatly as possible.  I also wonder if the pressure during atomization would damage the peptide.

I believe it's very important to have the reconstituting agent at the same temp as the peptide, whether it's Acetic Acid, Bac water or Sodium Chloride.  This will reduce any shock to the peptide becasue of difference in temps.

Hello everyone,
I'm with a documentary film production company, and we're currently researching a film on libido-enhancing pharmaceuticals.  I've been reading up on your posts about Bremelanotide usage, and I was hoping I could speak with any or all of you one on one about your experiences with it.  
We are particularly interested in speaking with women who have used it, so I was really interested in Miss Perfection's posts, and would love to chat with her (and any other women on this forum) if she's interested at all.  But in general, we want to speak to anyone about this "up and coming" pharmaceutical.

It is a protein and your saliva and stomach acid and enzymes will break it apart in the blink of an eye.
NOTHING with make it to the blood stream in any kind of active form.
